以對(duì)溴溴芐和亞磷酸三乙酯為原料合成4-溴芐基膦酸二乙酯(16)的一般步驟:在圓底燒瓶中加入1-溴-4-(溴甲基)苯(5.0 g,20 mmol)和亞磷酸三乙酯(51 mL,300 mmol),混合物在90℃下回流反應(yīng)19小時(shí)。反應(yīng)完成后,通過減壓蒸餾除去過量的亞磷酸三乙酯。粗產(chǎn)物通過快速柱色譜法(洗脫劑比例為1:1的己烷/乙酸乙酯)進(jìn)行純化,得到目標(biāo)化合物16,產(chǎn)率為98%。產(chǎn)物為無色液體;1H NMR(400 MHz,CDCl3)δ 7.30(d,2H,J = 7.5 Hz),7.05(d,2H,J = 7.6 Hz),3.99-3.88(m,4H),2.99(s,1H),2.94(s,1H),1.12(t,6H,J = 6.9 Hz);13C NMR(100 MHz,CDCl3)δ 131.7, 131.6, 131.5, 121.0, 62.3, 34.0, 32.0, 16.5;HRMS計(jì)算值(C11H16BrO3P,[M + H]+)307.0097,實(shí)測(cè)值307.0093。
