以噻唑-5-甲酸乙酯為原料合成5-羥甲基噻唑的一般步驟如下:在預(yù)冷(冰浴)的500mL三頸燒瓶中加入氫化鋁鋰(2.89g,76mmol)的250mL THF溶液。隨后,將噻唑-5-羧酸乙酯(11.82g,75.68mmol)溶于100mL THF中,并在1.5小時(shí)內(nèi)緩慢滴加到反應(yīng)體系中,以避免劇烈發(fā)泡。滴加完畢后,繼續(xù)攪拌反應(yīng)混合物1小時(shí)。反應(yīng)完成后,依次小心加入2.9mL水、2.9mL 15% NaOH溶液和8.7mL水以淬滅反應(yīng)。過濾去除固體鹽,保留濾液。將粗產(chǎn)物鹽在100mL乙酸乙酯中回流加熱30分鐘,然后過濾。合并兩次過濾所得的濾液,用無水Na2SO4干燥,隨后在減壓下濃縮。通過硅膠柱色譜法純化產(chǎn)物,依次使用0%、2%、4%甲醇的氯仿溶液作為洗脫劑,收集Rf值為0.3(4%甲醇的氯仿溶液)的餾分,靜置后固化,得到目標(biāo)化合物,收率為75%。產(chǎn)物結(jié)構(gòu)經(jīng)NMR(CDCl3)δ4.92(s,2H),7.78(s,1H),8.77(s,1H)和質(zhì)譜((M + H)+ = 116)確認(rèn)。
