以1-萘乙酮為原料合成1-萘乙胺的一般步驟:將相應(yīng)的酮(10mmol,具體為1.14g的1a,0.98g的1b,1.48g的1c,1.46g的1d,1.46g的1e,1.32g的1f,1.20g的1g,1.70g的1h,1.70g的1i)與羥胺鹽酸鹽(15mmol,1.04g)、甲酸銨(60mmol,3.78g)和鋅粉(30mmol,1.96g)在甲醇(30mL)中混合,于回流條件下攪拌反應(yīng)。反應(yīng)完成后,將反應(yīng)混合物通過Celite過濾。濾液經(jīng)真空旋轉(zhuǎn)蒸發(fā)除去溶劑。殘余物用濃鹽酸處理,隨后加入4mL HCl溶液和30mL水,用乙醚(2×20mL)萃取以去除有機(jī)雜質(zhì)。水相用氨水調(diào)節(jié)至pH=10,再用二氯甲烷(4×25mL)萃取。合并有機(jī)相,用飽和食鹽水洗滌,無水硫酸鈉干燥,最后真空除去溶劑得到目標(biāo)產(chǎn)物1-萘乙胺。
