以丁二酸酐和甲苯為原料合成3-(4-甲基苯甲酰)丙酸的一般步驟如下:將75.7 g丁二酸酐溶解于457.5 mL甲苯中。在攪拌下,分批加入220.5 g無水三氯化鋁(AlCl3)。維持反應(yīng)混合物溫度在50-60℃范圍內(nèi)。反應(yīng)2小時后,通過薄層色譜(TLC)監(jiān)測反應(yīng)完成情況。在劇烈攪拌下,將反應(yīng)混合物緩慢滴加至1250 mL 1 mol/L的冰鹽酸中。滴加過程中,逐漸有乳狀固體析出。滴加完畢后,繼續(xù)攪拌混合物0.5小時,隨后進(jìn)行過濾。濾餅依次用甲苯和冰水洗滌,然后用50 mL乙酸乙酯進(jìn)行抽濾。將所得溶液緩慢加入500 mL石油醚中,誘導(dǎo)結(jié)晶。攪拌0.5小時后,過濾收集晶體,并于40℃下干燥2小時,得到144 g化合物II(中間體),產(chǎn)率為99.3%。[化合物II中間體]為白色固體。
