以4-苯甲氧基苯胺鹽酸鹽為原料合成4-芐氧基苯肼鹽酸鹽的一般步驟:(4-(芐氧基)苯基)肼鹽酸鹽(2)。注意:在此過程中,將反應(yīng)混合物和所有添加的溶液保持在0℃。向反應(yīng)瓶中加入4-芐氧基苯胺鹽酸鹽(3.00g,12.5mmol)和水(25mL),在0℃下攪拌10分鐘。隨后,在15分鐘內(nèi)緩慢滴加NaNO2(852mg,12.3mmol)的水(6mL)溶液。滴加完畢后,繼續(xù)攪拌反應(yīng)混合物15分鐘。然后,緩慢加入SnCl2(6.40g,33.1mmol)的水(7.5mL)溶液。加完后,將反應(yīng)混合物在0℃下攪拌1小時(shí)。反應(yīng)完成后,過濾收集灰白色沉淀,用水洗滌,并用乙醚(Et2O)研磨,得到目標(biāo)產(chǎn)物2(3.01g,收率96%)。產(chǎn)物表征數(shù)據(jù)如下:IR(ATR,純)νmax 3232, 2906(br), 2693, 1568, 1508, 1242, 1177 cm-1;1H NMR(DMSO-d6, 600MHz)δ 10.11(bs, 3H), 7.44-7.40(m, 2H), 7.40-7.35(m, 2H), 7.33-7.29(m, 1H), 7.01-6.93(m, 4H), 5.05(s, 2H);13C NMR(DMSO-d6, 150MHz)δ 153.7, 139.1, 137.3, 128.5, 128.4, 128.3, 127.9, 127.7, 127.5, 117.1, 116.9, 115.5, 115.3, 69.5;HRMS(EI)m/z 計(jì)算值C13H14N2 214.1106,實(shí)測(cè)值214.1110。
