3.2 5-氨基-4-氰基-1-苯基-1H-吡唑(3a)的合成 將苯肼(2d)(0.45 mL,4.2 mmol)與乙氧基亞甲基丙二腈(1b)(0.51 g,4.2 mmol)溶于乙醇(4 mL)中。使用CEM Discover微波合成儀,在密封的額定壓力Pyrex管(10 mL)中,初始功率設(shè)置為100 W,于120℃下微波照射45分鐘。反應(yīng)完成后,在壓縮空氣流中冷卻,隨后通過減壓蒸餾除去溶劑。產(chǎn)物通過硅膠柱色譜純化,洗脫劑為石油醚/乙酸乙酯(5:1 v/v),得到1-苯基-4-氰基-5-氨基吡唑(0.738 g,收率95%),為淺棕色固體,熔點137-139℃(文獻(xiàn)值:137℃)。高分辨質(zhì)譜(實測值:185.0827,C10H9N4 [MH]+計算值:185.0822);傅里葉變換紅外光譜(KBr,cm?1)νmax:3302(NH),3240(NH),2230(CN),1577,1530,1368;1H NMR(400 MHz,CDCl3)δ:7.58(1H,s,H-3),7.49-7.37(5H,m,Ph),4.52(2H,br s,交換D2O后消失,NH2);13C NMR(100 MHz,CDCl3)δ:141.9(CH),135.0(C),129.3(CH),125.5(CH),124.1(CH),121.3(CH),115.6(C),86.2(C);低分辨質(zhì)譜(APcI)m/z(相對強度):226([M + MeCNH]+,100),185([MH]+,64)。
