以5-甲基-2-硝基芐腈(1.92g,11.84mmol)為原料,分批加入攪拌中的SnCl2(11.22g,59.2mmol)的濃鹽酸(12mL)和乙醇(EtOH,12mL)混合溶液中。反應(yīng)過程中使用冰浴控制溫度在20-30℃。反應(yīng)混合物在室溫下繼續(xù)攪拌1小時(shí)后,倒入預(yù)冷的6N NaOH水溶液(約30mL)中,中和至pH7。用乙酸乙酯(EtOAc)萃取產(chǎn)物,隨后用鹽水洗滌有機(jī)層,無水硫酸鎂(MgSO4)干燥,濃縮后得到目標(biāo)產(chǎn)物2-氨基-5-甲基苯腈(1.56g,收率99%),為黃棕色固體。產(chǎn)物結(jié)構(gòu)經(jīng)1H NMR(400MHz,DMSO-d6)δ2.21(s,3H),5.79(bs,2H),6.68-6.71(d,1H),7.10-7.13(dd,1H),7.15(s,1H);13C NMR(DMSO-d6)δ20.13,93.99,116.12,118.94,125.38,132.32,135.76,150.21;質(zhì)譜(MS)m/z 133(MH+)確認(rèn)。
