一般步驟:以2-(羥甲基)丙烯酸甲酯為原料合成(-)-甲基-D-BETA-羥基異丁酸酯的一般程序如下:Ene-還原酶活性測定需在環(huán)境溫度下于厭氧條件(Coy Laboratory,Grass Lake,MI)中進(jìn)行。反應(yīng)原液的制備包括將各底物(具體見下文)溶解于50mM Tris-HCl緩沖液(pH7.5)中,并補(bǔ)充200μM NADP+、100mM葡萄糖及來自Thermoplasma acidophilum的葡萄糖脫氫酶(GDH)(IVTT反應(yīng)使用2個單位,純化酶反應(yīng)使用5個單位)。選擇單個底物濃度以確保達(dá)到vmax條件或最大溶解度。在這些底物濃度下,調(diào)整反應(yīng)時間以達(dá)到10-50%的底物轉(zhuǎn)化率。對于IVTT實(shí)驗(yàn)中的OYE活性測定,將20μL反應(yīng)原液與2-10μL IVTT反應(yīng)混合物混合,并用50mM Tris-HCl(pH7.5)將總測定體積調(diào)整至30μL。對于純化的OYE1變體的測定,將酶(最終濃度:250nM)加入至500μL反應(yīng)儲備溶液中。通過從測定溶液中取出30μL等分試樣,并與30μL含有1mM環(huán)己酮(作為內(nèi)標(biāo))的乙酸乙酯充分混合以淬滅反應(yīng)進(jìn)程。收集有機(jī)相樣品并通過氣相色譜(GC)進(jìn)行分析(具體方案見下文)。通過積分產(chǎn)物和底物的峰面積來計(jì)算對映體/非對映異構(gòu)體過量。各底物的相對轉(zhuǎn)化率通過將測得的OYE1變體的轉(zhuǎn)化率除以野生型OYE1的相應(yīng)速率來計(jì)算。
