以咪唑和對(duì)氟苯甲醛為原料合成4-(咪唑-1-基)苯甲醛的一般步驟:在干燥條件下,將對(duì)氟苯甲醛(25.0g,0.200mol)、咪唑(0.300mol)和無(wú)水碳酸鉀(40.0g)的混合物溶于DMF(300mL)中,加入催化量的Aliquat 336試劑。將反應(yīng)混合物在100℃下回流24小時(shí)。反應(yīng)完成后,在減壓下濃縮反應(yīng)混合物并冷卻至室溫。隨后,將濃縮物倒入冰水中,靜置過(guò)夜。過(guò)濾析出的固體產(chǎn)物,用水洗滌,并用甲醇進(jìn)行重結(jié)晶,得到目標(biāo)化合物4-(1H-咪唑-1-基)苯甲醛(Ia),產(chǎn)率為92%,為黃色晶體,熔點(diǎn)為152℃(與文獻(xiàn)報(bào)道一致)[34,44]。
